چکیده
Title of Document: INHIBITORS OF AUTOINDUCER-2 QUORUM SENSING AND THEIR EFFECT ON BACTERIAL BIOFILM FORMATION Rebecca Melissa Lennen, Master of Science, 2007 Directed by: Dr. William Bentley, Professor Department of Bioengineering Bacteria utilize small signaling molecules, or autoinducers, to regulate their gene expression in tandem by a process termed quorum sensing. The gene encoding the synthase for autoinducer-2 (AI-2), luxS, is conserved in dozens of diverse bacteria. Behaviors controlled by AI-2 include virulence, motility, toxin production, and biofilm formation. The development of therapies that interfere with AI-2 quorum sensing are attractive for targeting biofilms, which exhibit inherent resistance to most antibiotics and biocidal agents. In this study, in vitro synthesized AI-2, LuxS inhibitors, and (5Z)-4bromo-5-(bromomethylene)-3-butyl-2(5H)-furanone were screened for their effect on biofilm formation in Escherichia coli, Bacillus cereus, and Listeria innocua. The LuxS inhibitors were found to have no influence on biofilm formation in any of the screened species, but reduced exponential phase AI-2 production in Listeria innocua. The brominated furanone significantly inhibited growth in B. cereus and L. innocua, and under certain conditions preferentially inhibited biofilm formation independently from

Transition state analogues of 5′-methylthioadenosine nucleosidase disrupt quorum sensing
5'-Methylthioadenosine/S-adenosylhomocysteine nucleosidase (MTAN) is a bacterial enzyme involved in S-adenosylmethionine-related quorum sensing pathways that induce bacterial pathogenesis factors. Transition state analogs MT-DADMe-Immucillin-A, EtT-DADMe-Immucillin-A and BuT-DADMe-Immucillin-A are slow-onset, tight-binding inhibitors of Vibrio cholerae MTAN (VcMTAN), with equilibrium dissociati...
متن کاملOpposite effects of cefoperazone and ceftazidime on S-ribosylhomocysteine lyase/autoinducer-2 quorum sensing and biofilm formation by an Escherichia coli clinical isolate
To investigate the effects of subminimum inhibitory concentrations of cephalosporins on bacterial biofilm formation, the biofilm production of 52 Escherichia (E.) coli strains was examined following treatment with cephalosporin compounds at 1/4 minimum inhibitory concentrations (MICs). Ceftazidime (CAZ) inhibited biofilm formation in seven isolates, while cefoperazone (CFP) enhanced biofilm for...
متن کاملRole of the luxS quorum-sensing system in biofilm formation and virulence of Staphylococcus epidermidis.
Nosocomial infections caused by Staphylococcus epidermidis are characterized by biofilm formation on implanted medical devices. Quorum-sensing regulation plays a major role in the biofilm development of many bacterial pathogens. Here, we describe luxS, a quorum-sensing system in staphylococci that has a significant impact on biofilm development and virulence. We constructed an isogenic Deltalux...
متن کاملChemotaxis towards autoinducer 2 mediates autoaggregation in Escherichia coli
Bacteria communicate by producing and sensing extracellular signal molecules called autoinducers. Such intercellular signalling, known as quorum sensing, allows bacteria to coordinate and synchronize behavioural responses at high cell densities. Autoinducer 2 (AI-2) is the only known quorum-sensing molecule produced by Escherichia coli but its physiological role remains elusive, although it is ...
متن کاملLow concentrations of local honey modulate ETA expression, and quorum sensing related virulence in drug-resistant Pseudomonas aeruginosa recovered from infected burn wounds
Objective(s): Honey’s ability to kill microorganisms and even eradication of chronic infections with drug-resistant pathogens has been documented by numerous studies. The present study is focused on the action of honey in its sub-inhibitory levels to impact on the pathogens coordinated behaviors rather than killing them. Materials and Methods:</strong...
